LightScanner® Master Mixes
LightScanner master mixes brilliantly provides a pre-optimized solution specifically designed to deliver superior PCR performance for
Hi-Res Melting and genotyping applications.
These master mixes reduce time and saves money
with its easy-to-use premixed formula.
It secures downstream success by directly going from PCR to melting curve analysis and eliminates the risk of contamination.
Idaho Technology offers three types of master mixes. See below for kit details.

This master mix incorporates a Taq polymerase and an antibody-based hot-start method, dNTPs, magnesium chloride and Idaho Technology's proprietary LCGreen® PLUS dye and is formulated at a 2.5X concentration. The antibody-mediated hot-start method means no enzyme pre-activation is required prior to or during thermocycling, translating into much faster PCR protocol times than other methods of hot-start protection.
The master mix includes a concentration of magnesium that is optimized to work for almost all reactions. Additional magnesium chloride solution is provided for further optimization if required. The master mix is also compatible with most additives used to enhance amplification efficiency of high GC amplicons. |

| High Sensitivity Master Mix Kit Contents |
This High Sensitivity
Master Mix is a pre-optimized master mix designed for
Hi-Res Melting® and subsequent genotyping of small amplicons.
This kit is specifically designed for mutation genotyping on the
LightScanner instrument.
This master mix incorporates specially tested Taq polymerase and
an antibody-based hot-start method, dNTPs, magnesium chloride,
and Idaho Technology’s proprietary LCGreen® Plus dye. It is formulated
at a 2.5X concentration. The antibody hot-start method means
that no enzyme pre-activation is required before thermocycling. In
addition, it includes special nucleic acid-based internal calibrators at
optimal concentrations to provide adequate signal when used with
amplicons up to 120 bp in size.
The master mix includes a universal magnesium buffer designed to work for almost all reactions. Additional magnesium chloride solution is provided for further optimization, if required. The master mix is also compatible with most additives used to enhance amplification efficiency of high G-C amplicons, although some additives may diminish hot-start efficacy. In general, only use additives, like DMSO or betaine, when necessary.

| Genotyping Master Mix Kit Contents |
This Genotyping Master Mix is a pre-optimized
master mix specially designed for genotyping experiments not
requiring an enzyme with exonuclease activity. This master mix is
compatible with technologies like SimpleProbes®, where the genotype
is determined via melting temperature of the SimpleProbe. This
product is intended for laboratory use only.
This master mix includes Taq polymerase, magnesium chloride, and bovine serum albumin (BSA) and is formulated at a 2.5X concentration. Enzyme preactivation (i.e., high temperature, hot start, incubation) is not required before thermocycling.
This master mix has been tested on all Idaho Technology real-time PCR instruments, Roche LightCycler®, and many commercially available heat block thermal cyclers. |
